Tirzepatide works by activating both glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP) receptors, improving blood-sugar control. Approved in the US, EU, Canada, and Australia, it's indicated for enhancing blood-sugar control in type 2 diabetes patients, alongside diet and exercise, and for chronic weight management in conjunction with a reduced-calorie diet and increased physical activity.

Compared to semaglutide, another diabetes medication, tirzepatide showed minor improvements in glycated hemoglobin levels (2.01%–2.30% depending on dosage vs. 1.86% for semaglutide). Tirzepatide's dual agonist behavior at GIP and GLP-1 receptors has been shown to produce greater reductions in hyperglycemia than a selective GLP-1 receptor agonist. It has also been found effective in reducing insulin resistance, with a reduction of around 8% from baseline at a 10 mg dose, measured using HOMA2-IR (computed with fasting insulin)
